Components

• 50 mM Tris-HCl, pH 7.4
• 150 mM NaCl
• 1% NP-40 (or IGEPAL CA-630)
• 0.5% Sodium deoxycholate
• 0.1% SDS
• Ajoutez frais avant utilisation : 1 mM PMSF, inhibiteurs de protéase, inhibiteurs de phosphatase

Preparation

Dissolvez components in deionized water, adjust pH to 7.4, filter sterilize, store à 4°C. Ajoutez inhibitors just avant utilisation. Final volume: 1 L.

Tampon d'Échantillon Laemmli

Tampon d'échantillon pour SDS-PAGE

Components

• 125 mM Tris-HCl, pH 6.8
• 4% SDS
• 20% Glycerol
• 0.004% Bromophenol blue
• 10% 2-mercaptoéthanol (ajoutez frais, ou utilisez 100 mM DTT)

Preparation

Mélangez components (except reducing agent), store à température ambiante. Ajoutez reducing agent just avant utilisation. For 2X buffer, mix 1:1 with sample. For reducing conditions, always add frais 2-ME or DTT.

Tampon de Transfert

Tampon pour le transfert de protéines

Components

• 25 mM Tris base
• 192 mM Glycine
• 20% Méthanol (pour les membranes PVDF)
• 0,1% SDS (optionnel, pour les grandes protéines >100 kDa)

Preparation

Dissolvez 3.03 g Tris base and 14.4 g glycine in 800 mL deionized water, add 200 mL methanol, adjust pH to 8.3, bring to 1 L. Stockez à 4°C. For nitrocellulose, reduce methanol to 10-15%.

TBST (Solution Saline Tamponnée Tris avec Tween-20)

Tampon de lavage

Components

• 20 mM Tris-HCl, pH 7.6
• 150 mM NaCl
• 0.1% Tween-20

Preparation

Dissolvez 2.42 g Tris base and 8.77 g NaCl in 900 mL deionized water, adjust pH to 7.6 with HCl, add 1 mL Tween-20, bring to 1 L, mix well. Stockez à température ambiante.

5% Lait Écrémé dans TBST

Solution de blocage standard pour la plupart des applications de western blot

Preparation

Dissolvez 5 g non-fat dry milk powder in 100 mL TBST. Mélangez well, may need to filter through cheesecloth to remove undissolved particles. Préparez frais or store à 4°C jusqu'à 1 week. Shake well avant utilisation.

3-5% BSA dans TBST

Solution de blocage pour les protéines phosphorylées ou lorsque le lait cause un bruit de fond élevé

Preparation

Dissolvez 3-5 g BSA (bovine serum albumin, Fraction V) in 100 mL TBST. Mélangez gently to avoid foaming. Filtrez through 0.45 μm filter if necessary. Stockez à 4°C. Utilisez within 1 week.

Solution de Coloration Ponceau S

Pour visualiser le transfert de protéines sur la membrane

Components

• 0.1% Ponceau S
• 5% Acetic acid

Preparation

Dissolvez 0.1 g Ponceau S in 100 mL of 5% acetic acid. Filtrez avant utilisation. Stockez à température ambiante. Stains membrane for 2-5 minutes, rinse with water to visualize bands.
